The M181 will be fully closed in both directions over the weekend while construction work begins on a new roundabout junction.
A diversion route will be in place during the closure, which will take place between Friday 25 October at 20:00 and Monday 28 October at 06:00.
The work between Frodingham and the M180 is part of the development of a new southern roundabout junction, which local authorities say will reduce congestion and reduce commute times to and from Scunthorpe.
Once it reopens the M181 will be reduced to one lane in each direction as works continue.
Work on the route first began in July and is set to be fully completed by Spring 2025.
